ENERGY METABOLISM OF BRAIN MITOCHONDRIA UPON OXIDATIVE STRESS AND IN AGEING

Background:
It is generally accepted that excessive production of ROS is critical for the decline of physiological functions that occurs during the ageing process. The mechanism of ROS production is not fully understood and there is still open debate concerning physiological consequences of the altered rates of ROS production in old animals. Moreover, our recently published article in Science (2007) identified a novel, unexpected pathway involved in ROS production mediated by PKCbeta which can be involved in ageing process.
Aim of the study:
The main goal of the project is to study correlations between age-dependent oxidative stress and cellular metabolism (in particular calcium homeostasis and bioenergetics of brain mitochondria).
International exchange:
It is anticipated that the PhD student will spend 24 months in Paolo Pintonís laboratory in the Department of Experimental and Diagnostic Medicine, University of Ferrara, Ferrara, Italy. During the stay the student will be trained in the technique of calcium measurement in different cellular compartments (cytosol, mitochondria and endoplasmic reticulum) with the use of aequorin - a calcium sensitive protein probe. The student will also learn microscopic and confocal techniques.
Additional requirements for the student:
The candidate should have master degree in biological sciences within 2- 3 years before the call. Good command of English is required.
