REGULATION OF FUNCTION AND LOCALIZATION OF CARNITINE TRANSPORTERS IN THE BLOOD-BRAIN BARRIER

Background:
Blood-brain barrier, formed by the brain capillary endothelial cells, controls uptake and removal of many important nutrients and other low molecular weight compounds, due to an often polarized localization of specific transporting proteins. L-carnitine is necessary in catabolism of fatty acids, mainly in transfer of acyl moieties to mitochondrial matrix and export of medium-chain acylcarnitines from peroxisomes. Carnitine can be transported to the cell by proteins belonging to OCTN family (Organic Cation Transporters Novel family) or by an amino acid transporter B0,+. Apart from their important physiological role, carnitine transporters have not been studied till now, as regards their regulation at post-translational level.
Aim of the study:
The project is aimed at studies of transporting activity of carnitine transporters and their localization within the cells forming the blood-brain barrier under conditions influencing protein phosphorylation.
International exchange:
The PhD student will spend 12 months abroad (three visits, 4 months each) in the laboratory of prof. R. Cecchelli (Université d’Artois, Faculté des Sciences Jean Perrin, Lens, France). He/she will learn the technique of culturing brain capillary endothelial cells in a co-culture with glial cells, followed by transport measurements, immunoprecipitation experiments and preparing cells for immunocytochemistry experiments.
Additional requirements for the student:
Education: MSc, specialized in cell biology, with very good education in biochemistry and scientific interest in protein chemistry and biological membranes, fluent English; Skills: laboratory experience (apprenticeship during university studies); Additional: Good health conditions in order to get permission for work with the radioactive substances
