THE ROLE OF PROTEIN ARGINYLATION IN CYTOSKELETON REMODELLING IN NEURONAL CELLS IN PHYSIOLOGY AND PATHOLOGY

Background:
Protein arginylation, a post-translational modification mediated by arginyltransferase (Ate1), was originally believed to induce degradation of the target protein substrates by the ubiquitin-dependent N-end rule pathway. However, it has been recently found that arginylation has a much broader spectrum of targets and more complex effects in vivo which remain to be fully understood. Protein arginylation may regulate different proteins in distinct ways affecting their assembly and structure, metabolic stability and/or other properties that facilitate their intracellular functions in many important biological processes.
Aim of the study:
The main aim of this PhD project is to establish the role of actin and tubulin arginylation in neuronal plasticity.
International exchange:
The PhD student is expected to visit the Laboratory of Cell Signaling of the Institute of Cell Biology NASU for four 3-month-long visits to purify arginine catabolizing enzymes and to use them to obtain arginine-deprived medium to culture the studied cell lines. Also, the student will perform the arginyltransferase (Ate1) knockdown by means of siRNA technique, as well as monitor autophagic process upon different experimental conditions in neuronal cells.
Additional requirements for the student:
The candidate should possess a master degree in biological sciences earned within three years before the call; graduates of medical and veterinary schools are also eligible to apply. The candidate is expected to have a strong scientific interests and ability to work in a team. Good command of English and two letters of recommendation are required.
